The essential element of course, to any backpacking trip is a good backpack. It doesn't have to the latest and most expensive back pack available, but it does have to be hard-wearing, and contain all the compartments you might need. Most back packs have capacities measured in litres - and a lot of backpacker tips mention going for a 65 litre backpack.
Of course, it really depends on how long you are going for, but for a year long trip, you would be best advised to have at least 65 litres. And it depends how many creature comforts you want and how often you plan to wash your clothes. It is true that once backpackers get deep into their trip, their clothes do indeed get washed less - often being cleaned in the shower on even in a river, depending on surroundings. Insect repellent (mosquito spray), sun tan lotion and a basic medical kits of cuts and bruises - these are all very advisable, although most people would find they never use the first aid kit. However fortune favours the prepared, or something like that! Readiness is all, said Shakespeare.

Depends on where you are going. In general, pack light. Think layers. Socks and shoes are the key to comfort. But without knowing the climate, this is an impossible question to answer fully.
